All decent tuxedo shirts should have French cuffs, which have holes for cuff links. You can ask at the rental shop, but it would be a low-class place indeed that rents formal shirts with buttoning cuffs.

I've always loved French cuffs. They're twice as long as the ordinary cuff, and have four holes--you fold the cuff over itself, and slip the link through all four.

Now, with a tuxedo, you would traditionally go for more formal cuff links--gold or silver with onyx inlay, for instance. That said, weddings are a time for frivolity, and cuff links are one of the more tasteful ways of having fun with a tuxedo. (Long ties, on the other hand, are not a tasteful way of having fun with a tuxedo.)
References :
You can call the place they are renting the tux from and make sure that he gets a shirt that has holes for cuffs. If you want it to be a total surprise, see if the tux rental store will take the cuff links from you and present them to him when he picks up a the tux.

My husband buys shirts that have buttons AND have discrete dual holes for cuff links for more formal/dressy occasions.

Congrats and best wishes for your wedding and future together!
References :
not all white dress shirts have a cufflink hole. YOu can always buy a plain white french cuff(uses cufflinks) shirt for $20 at the low end
